反相高效液相色谱法测定复方大黄颗粒中大黄素和大黄酚的含量

     摘要:目的建立反相高效液相色谱(RPHPLC)法测定复方大黄颗粒中大黄素和大黄酚的含量。方法Alltima C18色谱柱(5 μm,150 mm×4.6 mm);柱温:30℃;流动相:甲醇0.1%磷酸(87∶13);流速:1 ml/min;检测波长:254 nm。结果大黄素的线性范围为0.096~0.576 μg(r=0.999 9),平均回收率为99.29%,RSD为0.63%;大黄酚的线性范围为0.078 4~0.470 4 μg(r=0.999 8),平均回收率为100.08%,RSD为1.52%。结论该方法灵敏度高、操作简便、重现性好、效率高。可作为样品的检测方法。

    Determination of Emodin and Chrysoplhanol in Compound Rhubard Granula by RPHPLC

    Abstract:ObjectiveTo develop an RPHPLC method for determination of emodin and chrysophanol in Compound Rhubard Granula.MethodsThe samples were determined by RPHPLC with a Alltima C18column(5 μm,150 mm×4.6 mm) and UV detector at 254 nm.The mobile phase was methonal∶0.1%H3PO4(87∶13)at a flow rate of 1.0 ml/min.The column temperature was 30℃.ResultsThe calibration curve of emodin was linear over the ranges of 0.096μg~0.576 μg (r=0.999 9),average recovery was 99.29,RSD was 0.63%;The calibration curve of chrysophanol was linear over the ranges of 0.078 4~0.470 4 μg (r=0.999 8),average recovery was 100.08%,RSDwas 1.52%.ConclusionThis method is simple,reliable,efficient,and it provides a reliable method for determination of emodin and chrysophanol in Compound Rhubard Granula. ......
